Propath offers a complete, turnkey service for GLP and GCP-compliant IHC staining, using automated and manual methods.
The team at Propath has substantial experience of developing and validating robust methods for experimental and regulated studies.
To generate meaningful study data that can be relied upon, we appreciate the importance of producing staining results that are consistent, sensitive and reproducible.
IHC is a complex, multi-parameter staining technique applied across variable fresh frozen and fixed paraffin embedded tissues. As a result, developing an optimum staining protocol is not always straightforward.
Our experience informs our view that high quality IHC with strong signal intensity and low background staining can only be achieved by paying careful attention to the following factors:
Whether developing an IHC method for a proprietary antibody clone or staining specimens from a large scale clinical trial; Propath has the experience and technology platforms to ensure your study is as successful as possible.
For routine IHC, we use the Roche Ventana Discovery Ultra and Leica Bond Rx III– both platforms well recognised for providing high quality, consistent and reproducible staining outcomes.
For more advanced multiplexed immunofluorescence, we offer access to the powerful Lunaphore Comet and Akoya PhenoCycler-Fusion and Pheno-imager platforms.
All IHC is conducted onsite by experienced scientists within our fully GLP and GCP compliant environment, providing Propath clients further confidence in our standards of quality assurance.
The team at Propath offer significant experience in developing and validating IHC and ISH assays for clinical, bio distribution, tissue cross-reactivity, efficacy, pharmacodynamics and toxicity studies.
We have worked with a broad range of biomarkers and antibody structures, and developed a range of strategies to overcome common technical challenges.
We use the Ventana Discovery Ultra and Leica Bond Rx for routine IHC staining, which both provide consistent and reproducible staining. For high-plex staining, we use our Lunaphore COMET system, which offers up to 40-plex multiplexing per slide.
Propath validates new IHC methods using appropriate positive and negative control tissue to assess specificity, sensitivity, linearity, repeatability (precision) and reproducibility. Assay robustness is tested across different staining machines with different scientists performing the assay. Where required, different reagent lots are tested and the assay can be run on a panel of tissues/ TMAs to assess suitability of the assay across different sample types.
Using our Ventana Discovery Ultra and Leica Bond Rx platforms, we are able to develop methods for up to 4-plex per slide. In addition, using Ultivue panels or our Akoya staining platfom we are able to multiplex up 8-plex per slide.
Yes - using our Ventana or Leica autostainer platforms we can develop automated ISH methods, including RNAscope, multiplex ISH and combined ISH/IHC multiplex.
